Poster Abstract: Automating High-Efficiency Genomic Workflows: Agilent SureSelect Max DNA Library Preparation and Target Enrichment on the firefly® Liquid Handling Platform

Abstract

Next-generation sequencing core facilities demand scalable, walkaway solutions to meet growing genomic data requirements. We have automated the Agilent SureSelect Max DNA Library Preparation with Enzymatic Fragmentation and Target Enrichment using Fast Hybridization on the SPT Labtech firefly+® liquid handling platform, delivering a streamlined, end-to-end workflow with minimal hands-on time.
 

Leveraging the firefly+ on-deck thermocycler, the entire process from enzymatic fragmentation to probe hybridization runs on a single deck, supporting 1 to 96 samples for library preparation and flexible pooling configurations for target enrichment.
 

Validation using 200 ng of genomic DNA demonstrated exceptional reproducibility across a 96-well plate (341 bp average fragment size, 3.0% CV; 4,490 nM average yield, 9.5% CV). Post-capture sequencing on the Element AVITI™ showed on-target rates above 80%, 30X base coverage above 93% for both exome V8 and Cancer CGP panels, and variant calling accuracy at or above 99%, even for pre-capture pooled libraries.
